    System Design of an Autonomous Underwater Robot “DaryaBird”

    Various kinds of robots have been developed parallel with the progress of computers and information processing technology, and the operations in the extreme environments, such as disaster areas, space and ocean, are getting one of the practical solutions for those hazardous missions. The underwater robots are one of the extreme environment robots and expected as one of solutions for underwater activities i.e., maintenance of underwater structures, observations, scientific research, where research area is getting wide and deep and also underwater structures are getting large-scale and deep-depth. Their efficiencies have been investigated during recent decades and are proven by ocean experiments. However, the robotic system including the support vessels is still big scale, and not so easy to handle by a few researchers. In this paper, we describe the design of an underwater robot “DaryaBird” developed aiming at handy, small underwater robots which can be operated by a few researchers.     Endothelial Progenitor Cells Promote Directional Three-Dimensional Endothelial Network Formation by Secreting Vascular Endothelial Growth Factor

    Endothelial progenitor cell (EPC) transplantation induces the formation of new blood-vessel networks to supply nutrients and oxygen, and is feasible for the treatment of ischemia and cardiovascular diseases. However, the role of EPCs as a source of proangiogenic cytokines and consequent generators of an extracellular growth factor microenvironment in three-dimensional (3D) microvessel formation is not fully understood. We focused on the contribution of EPCs as a source of proangiogenic cytokines on 3D microvessel formation using an in vitro 3D network model. To create a 3D network model, EPCs isolated from rat bone marrow were sandwiched with double layers of collagen gel. Endothelial cells (ECs) were then cultured on top of the upper collagen gel layer. Quantitative analyses of EC network formation revealed that the length, number, and depth of the EC networks were significantly enhanced in a 3D model with ECs and EPCs compared to an EC monoculture. In addition, conditioned medium (CM) from the 3D model with ECs and EPCs promoted network formation compared to CM from an EC monoculture. We also confirmed that EPCs secreted vascular endothelial growth factor (VEGF). However, networks cultured with the CM were shallow and did not penetrate the collagen gel in great depth. Therefore, we conclude that EPCs contribute to 3D network formation at least through indirect incorporation by generating a local VEGF gradient. These results suggest that the location of EPCs is important for controlling directional 3D network formation in the field of tissue engineering

    Enamel Carious Lesion Development in Response to Sucrose and Fluoride Concentrations and to Time of Biofilm Formation: An Artificial-Mouth Study

    The aim of this study was to evaluate both sucrose and fluoride concentrations and time of biofilm formation on enamel carious lesions induced by an in vitro artificial-mouth caries model. For Study 1, biofilms formed by streptococci and lactobacilli were grown on the surface of human enamel slabs and exposed to artificial saliva containing 0.50 or 0.75 ppmF (22.5 h/d) and broth containing 3 or 5% sucrose (30 min; 3x/d) over 5 d. In Study 2, biofilms were grown in the presence of 0.75 ppmF and 3% sucrose over 3 and 9 days. Counts of viable cells on biofilms, lesion depth (LD), and the integrated mineral loss (IML) on enamel specimens were assessed at the end of the tested conditions. Counts of total viable cells and L. casei were affected by sucrose and fluoride concentrations as well as by time of biofilm formation. Enamel carious lesions were shallower and IML was lower in the presence of 0.75 ppmF than in the presence of 0.50 ppmF (P 0.25). Additionally, deeper lesions and higher IML were found after 9 d of biofilm formation (P < 0.005). Distinct sucrose concentrations did not affect enamel carious lesion development. The severity of enamel demineralization was reduced by the presence of the higher fluoride concentration. Additionally, an increase in the time of biofilm formation produced greater demineralization. Our results also suggest that the present model is suitable for studying aspects related to caries lesion development

    本研究は、小学校学級担任を対象に、学級運営等に関するDaily Hassles(日常苛立ち事)を測定する尺度の開発を目的とした。A県の公立小学校50校の通常学級担任663名を対象に、質問紙調査を実施した。項目反応理論を用いた内容的妥当性の検討、構造方程式モデリングによる因子構造ならびに外的基準との関係での構成概念妥当性の検討、並びに内的整合性に着目した信頼性の検討を行った。その結果、最終的に選定した児童関連Hassles 9項目、保護者関連Hassles 4項目で構成された小学校学級担任の「職務関連Daily Hassles測定尺度」(経験頻度・ストレス強度)の構成概念妥当性と信頼性は統計学的に支持された。本研究の結果は、今後の教員のストレスと教員自身が持つ内的・外的資源との関連性の検討やストレス更には精神的健康の悪化を防ぐための支援・予防策の検討にとって有意義な基礎資料となることが示唆された。The purpose of this research was to develop the Daily Hassles Scale for elementary school homeroom teachers\u27 class-management. We conducted a survey on 663 regular homeroom teachers of public elementary schools in A prefecture. We examined the content validity of the questionnaire by means of item response theory. We also examined its construct validity in a relation with the factor structure and the external criterion by means of SEM, and its reliability which paid its attention to inner consistency. The balidity and reliability of "Daily Hassles Scale for Class-Management" which we composed of 9 items for students\u27 affairs and 4 items for parents\u27 affairs were good enough. These findings suggested that using this hassles scale may be useful for examination of the relationship between teachers\u27 stress and their internal/external resources and the effective support to maintain teachers\u27 mental health

    Non-touch, Quick Removal of an Occluding Intratracheal Balloon Using High Intensity Focused Ultrasound and Limonene Emulsion

    In recent years, fetal endoscopic tracheal occlusion (FETO) using a balloon has been clinically employed for promoting prenatal lung growth to ameliorate postnatal respiratory failure caused by severe in utero lung hypoplasia. After a successful FETO, in some limited fetal centers, planned or emergency balloon removal using another fetoscopy is performed to release the tracheal occlusion immediately before delivery. To overcome this additional fetoscopy, we previously reported an innovative simple procedure to remove the occluding balloon by bursting it with a pre-planned sequence of high intensity focused ultrasound (HIFU) irradiation. In that previous study, which used rabbits euthanized and submerged in degassed water, we inflated the balloon by injecting a mixture of perfluorocarbon and ultrasound contrast medium through a fetoscopically-guided catheter. The rate of successful balloon burst and deflation using HIFU irradiation was high enough (100%), but the mode and timing of tracheal reopening (i.e., sudden burst or slow shrinkage of the balloon) was rather varied and collateral damage to the dermal/tracheal tissue was identified in 72.7% of the experimental animals. Accordingly, to standardize the HIFU irradiation sequence and to achieve a reliable and secure balloon burst, we conducted another series of animal experiments in which the mixture of perfluorocarbon was replaced with “limonene emulsion” (D-limonene micelle emulsified in physiological saline) as the balloon injection fluid. In all 6 animals, we succeeded in reopening the airway by achieving an instantaneous and timely balloon burst without definite skin/tracheal damage. We conclude that HIFU irradiation together with injecting the balloon with “limonene emulsion” is an improved method for safe tracheal release from a balloon occlusion

    The Tokyo Oldest Old Survey on Total Health (TOOTH): A longitudinal cohort study of multidimensional components of health and well-being

    <p>Abstract</p> <p>Background</p> <p>With the rapid worldwide increase in the oldest old population, considerable concern has arisen about the social and economic burden of diseases and disability in this age group. Understanding of multidimensional structure of health and its life-course trajectory is an essential prerequisite for effective health care delivery. Therefore, we organized an interdisciplinary research team consisting of geriatricians, dentists, psychologists, sociologists, and epidemiologists to conduct a longitudinal observational study.</p> <p>Methods/Design</p> <p>For the Tokyo Oldest Old Survey on Total Health (TOOTH) study, a random sample of inhabitants of the city of Tokyo, aged 85 years or older, was drawn from the basic city registry. The baseline comprehensive assessment consists of an in-home interview, a self-administered questionnaire, and a medical/dental examination. To perform a wide variety of biomedical measurements, including carotid ultrasonography and a detailed dental examination, participants were invited to our study center at Keio University Hospital. For those who were not able to visit the study center, we provided the option of a home-based examination, in which participants were simultaneously visited by a geriatrician and a dentist. Of 2875 eligible individuals, a total of 1152 people were recruited, of which 542 completed both the in-home interview and the medical/dental examination, with 442 completed the in-home interview only, and another 168 completed self or proxy-administered data collection only. Carotid ultrasonography was completed in 458 subjects, which was 99.6% of the clinic visitors (n = 460). Masticatory assessment using a colour-changeable chewing gum was completed in 421 subjects, a 91.5% of the clinic visitors.</p> <p>Discussion</p> <p>Our results demonstrated the feasibility of a new comprehensive study that incorporated non-invasive measurements of subclinical diseases and a detailed dental examination aiming at community-dwelling individuals aged 85 years or older. The bimodal recruitment strategy is critically important to capture a broad range of health profiles among the oldest old. Results form the TOOTH study will help develop new models of health promotion, which are expected to contribute to an improvement in lifelong health and well-being.</p> <p>Trial Registration</p> <p>This study has been registered in the UMIN-Clinical Trial Registry (CTR), ID: UMIN000001842.</p

    A genome-wide gain-of-function analysis of rice genes using the FOX-hunting system

To elucidate the functions of a large population of rice genes and to search efficiently for agriculturally useful genes, we have been taking advantage of the Full-length cDNA Over-eXpresser (FOX) gene-hunting system. This system is very useful for analyzing various gain-of-function phenotypes from large populations of transgenic plants overexpressing cDNAs of interest and others with unknown or important functions. We collected the plasmid DNAs of 13 980 independent full-length cDNA (FL-cDNA) clones to produce a FOX library by placing individual cDNAs under the control of the maize Ubiquitin-1 promoter. The FOX library was transformed into rice by Agrobacterium-mediated high-speed transformation. So far, we have generated approximately 12 000 FOX-rice lines. Genomic PCR analysis indicated that the average number of FL-cDNAs introduced into individual lines was 1.04. Sequencing analysis of the PCR fragments carrying FL-cDNAs from 8615 FOX-rice lines identified FL-cDNAs in 8225 lines, and a database search classified the cDNAs into 5462 independent ones. Approximately 16.6% of FOX-rice lines examined showed altered growth or morphological characteristics. Three super-dwarf mutants overexpressed a novel gibberellin 2-oxidase gene, confirming the importance of this system. We also show here the other morphological alterations caused by individual FL-cDNA expression. These dominant phenotypes should be valuable indicators for gene discovery and functional analysis.publishersversionPeer reviewe
